In other words, “I don’t know.” I’ve read a few biological protocols as a scientific lay person and they are filled with instructions that say to heat something at some temp for some number of minutes, but never explain why. The only reason why according to those in the know turns out to be “we tried several different times and temps and this was the most optimal for whatever reason” or “the physics is really complicated; just trust us on this”.
